Abstract

Pendahuluan: Kanker merupakan kelainan genetik dan metabolik yang menjadi tantangan yang besar pada abad ke-21 ini. Dewasa ini terbukti bahwa Reactive Oxygen Species (ROS) dapat memicu karsinogenesis melalui beberapa jalur ongkogenik atau mutasi ongkogenik pada DNA. Oleh sebab itu diperlukan antioksidan yang dapat meregulasi jumlah ROS dalam sel kanker. Selain itu, hampir semua aktivitas fitokimia bekerja dalam beberapa jalur pensinyalan sel dan dapat berpotensi menghambat progresi keganasan. Salah satu fitokimia yang dapat memicu apoptosis adalah alkaloid karbazol yang terdapat pada daun kari (Murraya koenigii L.). Penelitian ini dilakukan untuk mengetahui kandungan fitokimia, kapasitas antioksidan, dan sitotoksisitas ekstrak metanol daun kari.

Metode: Penelitian ini menggunakan ekstrak metanol daun kari yang diperoleh dengan metode perkolasi. Ekstrak kemudian dilakukan pengujian secara invitro untuk mengetahui kandungan fitokimia dan kapasitas antioksidan dengan metode 3-ethylbenzothiazoline-6-sulfonic acid (ABTS). Selain itu, dilakukan juga pengujian dengan bioassay pada uji sitotoksisitas metode Brine Shrimp Lethality Test (BSLT).

Hasil: Ekstrak metanol daun kari mengandung alkaloid, antosianin, koumarin, flavonoid, kuinon, saponin, steroid, tanin, dan terpenoid, dengan kapasitas antioksidan yang dinyatakan dalam IC50 sebesar 30,70 μg/mL, sedikit lebih lemah dibandingkan Trolox sebesar 16,46 µg/mL. Sedangkan uji sitotoksisitas pada ekstrak metanol daun kari didapatkan LC50 sebesar 235,34 µg/mL.

Simpulan: Ekstrak metanol daun kari memiliki beragam fitokimia dengan aktivitas antioksidan yang sangat kuat (> 150 µg/mL) dan tergolong toksik (31 – 1000 µg/mL) sehingga bersifat sitotoksik terhadap sel yang sedang membelah sehingga memiliki potensi sebagai antikanker.
